Design Notes
TetR was designed without a stop codon to enable the construction of a fusion protein with yeast codon optimized BFP. BFP was also designed without a stop codon for the purposes of integrating into the Ade2 locus of S. cerevisiae. This Ade2 locus contains a terminator region flanking BFP downstream.

The TetR repressor was PCR amplified from a plasmid source of the BioBrick (BBa_K319046) that was submitted last year by the uOttawa team. Yeast codon optimized BFP was synthesized by the uOttawa team using Mr. Gene DNA synthesis.
